One of the first and foremost test which the male partner needs to undergo is a semen analysis.  The semen analysis can show various abnormalities like no sperms, or a low count of sperms, low motile sperms and also low normal looking sperms. Based on which parameter is affected, the male partner is then asked to undergo certain blood tests or ultrasound or further specialized testing, as per need.  An examination by an andrologist may also be required in certain cases.

It is a condition where because of some underlying problem, the male is unable to get the wife pregnant. It could either be low sperm count (oligozoospermia), absence of sperms /no sperms (azoospermia), less motile sperms (asthenozoospermia), or abnormal looking sperms (teratozoospermia).
The male is asked about any symptoms like decreased sexual drive, decreased facial / body hair, any pain in genital/scrotal area, smoking, alcohol or drug intake, any sexual disturbances. If required a genital examination may be carried out followed by blood hormones if necessary.
Semen Analysis is essential to assess the sperm quantity and quality.
Additional tests in few cases may be required:
DNA fragmentation index (DFI)
Y chromosome microdeletion
Cystic fibrosis gene testing
Karyotyping
Sperm Freezing
Fertility with male factor is possible. The couple may require IUI or TESE (Testicular sperm extraction) followed by ICSI according to the problem and severity. In some cases, last resort may be a donor sperm.
